Transcriptomic Analysis of Differentially Expressed Genes in Arabidopsis thaliana Overexpressing BnMYB2 from Boehmeria nivea under Cadmium Stress

چکیده

Boehmeria nivea (ramie) is an important fiber crop with strong tolerance to cadmium (Cd). In our previous study, a novel MYB transcription factor gene from ramie, BnMYB2, was found positively regulate Cd and accumulation in the transgenic Arabidopsis thaliana lines. Herein, transcriptome sequencing performed identify differentially expressed genes involved response between wild-type (WT) BnMYB2 overexpressed lines; 1598 (DEGs) were detected shoot. GO KEGG analysis indicated that majority of DEGs belonged categories factors, plant hormone signal transduction nitrogen metabolism. The expression level Ib subgroup bHLH (AtbHLH38, AtbHLH39, AtbHLH100 AtbHLH101) assimilation-related (AtNIA1, AtNIA2, AtNIR1 AtASN2) significantly higher than WT, accompanied positive changes iron (Fe) total content shoot overexpression Several belonging bZIP family or SAUR also up-regulated plants. These results provide clues for elucidating how molecular mechanisms stress.

Development and validation of molecular markers for characterization of Boehmeria nivea var. nivea and Boehmeria nivea var. tenacissima

BACKGROUND The root of Boehmeria spp (ramie) is a hepatoprotective Chinese herbal medicine. Medicinal properties vary between Boehmeria nivea var. nivea and Boehmeria nivea var. tenacissima, which are local species found in Taiwan. As commercial preparations may use either species, there is a need for a rapid and simple assay to identify variants for quality control. METHODS Four methods were...

Identification of Differentially Expressed Genes in RNA-seq Data of Arabidopsis thaliana: A Compound Distribution Approach

Gene expression is the process by which information from a gene is used in the synthesis of a functional gene product, which may be proteins. A gene is declared differentially expressed if an observed difference or change in read counts or expression levels between two experimental conditions is statistically significant. To identify differentially expressed genes between two conditions, it is ...

Analysis of miRNA expression under stress in Arabidopsis thaliana.

MicroRNAs (miRNAs) are a class of small, 21-24 nucleotides long, non-coding RNAs involved in the post-transcriptional regulation of gene expression. Using the array analysis on Arabidopsis thaliana infected with the Oil-seed Rape Mosaic Virus (ORMV), we have found 28 up-regulated miRNAs. From them, six were selected for further validation by Northern blot analysis: miRNA172a, miRNA161, miRNA167...

Transcriptome Sequencing and Profiling of Expressed Genes in Phloem and Xylem of Ramie (Boehmeria nivea L. Gaud)

Ramie (Boehmeria nivea L. Gaud) is a highly versatile herbaceous plant which is widely cropped in southern China. The success of this herbaceous plant relies on wide use in modern industry. Understanding the profiling of expressed genes in phloem and xylem of ramie is crucial for improving its industrial performance. Herein, we uncover the transcriptome profile in phloem and xylem in present st...

Whole-transcriptome sequence analysis of differentially expressed genes in Phormium tenax under drought stress

Phormium tenax is a kind of drought resistant garden plant with its rich and colorful leaves. To clarify the molecular mechanism of drought resistance in Phormium tenax, transcriptome was sequenced by the Illumina sequencing technology under normal and drought stress, respectively. A large number of contigs, transcripts and unigenes were obtained. Among them, only 30,814 unigenes were annotated...
